Intestinal crypt homeostasis revealed at single-stem-cell level by in vivo live imaging

This paper addresses the identity and dynamics of intestinal stem cells in homeostasis. Using a combination of in vivo live-imaging and static clonal assays, we use biophysical modelling to show that maintenance involves a process of dynamical heterogeneity in which stem cells move reversibly between states primed for differentiation and primed for self-renewal. We show that the short-term potency of stem cells is correlated with position in the stem cell niche.
